This locus represents the 5\' regulatory region of the bone morphogenetic protein 2 gene, and contains both promoters and negative regulatory regions. A negative regulatory region downstream of the start of transcription may block activation of this promoter by a long-range downstream enhancer element. This region may be methylated and silenced in cancers. This locus also includes two accessible chromatin subregions that were validated as silencers based on their ability to repress an origin of replication minimal core promoter by the ATAC-STARR-seq (assay for transposase-accessible chromatin with self-transcribing active regulatory region sequencing) massively parallel reporter assay (MPRA) in GM12878 lymphoblastoid cells.
