This region includes the proximal promoter and nearby downstream enhancer of the SRY-box 9 (SOX9) gene. This promoter is regulated by signaling through interleukin 1 beta, notch, and transforming growth factor beta 1. Cytosine methylation may block transcription factor binding in this region, resulting in transcriptional silencing. This locus also includes two accessible chromatin subregions that were validated as silencers based on their ability to repress an origin of replication minimal core promoter by the ATAC-STARR-seq (assay for transposase-accessible chromatin with self-transcribing active regulatory region sequencing) massively parallel reporter assay (MPRA) in GM12878 lymphoblastoid cells.
