This genomic region includes the SOX2 regulatory region 2 (SRR2), a conserved sequence that functions as an enhancer. A subregion was shown to regulate the SOX2 (SRY-box 2) promoter located approximately 4 kb upstream by reporter assays in PA-1 human teratoma cells, where HMG and POU binding site sequences are important for function. The SRR2 enhancer additionally showed cooperative activity with the SRR1 enhancer located approximately 4 kb upstream of SOX2. This genomic region was also validated as an active enhancer by the ChIP-STARR-seq massively parallel reporter assay in primed human embryonic stem cells, where it associates with the OCT4 and NANOG transcription factors and is marked by the H3K27ac histone modification.
